    Evaluation of Trichoderma harzanium treated cassava waste on the quality and quantity of milk of goat

    The effects of the fungus, Trichoderma harzanium, treated cassava waste on milk quantity and quality were investigated in a completely randomized design model with twelve lactating West African dwarfdoes. Goats were fed the experimental diets containing O (A), 20 (B), 30 (C) and 40% (D) Trichoderma treated cassava waste during a 13 week lactation trial. The results revealed significant differences inmilk quantity and quality. The highest milk yield was recorded for diet D followed closely by diets C, B and A in that order. Inclusion of fungus treated cassava waste resulted in significant increasing milk fat,protein, solids not fat (SNF), and total solids. While the inclusion had little effect on milk pH and specific gravity. The yield of 40% fat corrected milk (FCM) and gross efficiency of milk increased significantly (P < 0.05) by the addition of fungus treated cassava waste in the diet. In conclusion, the addition of Trichoderma treated cassava waste was beneficial to lactating West African dwarf goats

    Effect of &#946-Glucuronidase on Extraction Efficiency of Silymarin from Human Plasma Samples Using Validated HPLC-UV Analysis

    Purpose: To investigate the effect of &beta;-glucuronidase on the extraction efficiency of silymarin (mainly as silybin) from spiked human plasma using a sensitive and reproducible high performance liquid chromatography (HPLC) method.Methods: The importance of &beta;-glucuronidase was evaluated by comparing the extraction efficiency of silymarin in &beta;-glucuronidase-treated and untreated plasma samples. Isocratic HPLC with simple UV detection (288 nm) was applied to analyze the major silymarin components using Thermo-Electron C18 column (200 mm, 4.6 mm I.D., 5&mu;m particle size). The mobile phase, consisting of methanol and 20 mM potassium dihydrogen phosphate buffer (50:50 v/v pH 2.8), was pumped at 1 ml/min.Results: The mean extraction efficiency was 98.97 % (CV = 1.69 %) for treated and 40.88 % (CV = 2.77 %) for untreated plasma samples, compared with nominal concentrations.Conclusion: The studied method showed 60 % reduced extraction efficiency of untreated samples compared to treated samples.Keywords: Silymarin, Silybin, Extraction Efficiency, &beta;-glucuronidase, HPL

    Budget impact analysis of rituximab biosimilar in Italy from the hospital and payer perspectives

    Introduction: This article aims at investigating the 5-year budget impact of rituximab biosimilars in Italy. Methods: A budget impact analysis model was developed in accordance with the International Society For Pharmacoeconomics and Outcomes Research recommendations. Drug acquisition and drug administration costs were considered since the risk/benefit profile of biosimilars and the originator was assumed to be overlapping. The perspectives of hospitals and payers were used. Input data were retrieved from the literature and validated/integrated by an expert panel of seven clinicians from various Italian regions. A dynamic incidence-based approach was used. Results: From the hospital perspective, adopting a rituximab biosimilar would produce savings of €79.2 and €153.6 million over 3 and 5 years, respectively. The results are very similar if the payer perspective is considered, with a cumulated savings of about €153.4 million in 5 years. Lymphoma and chronic lymphocytic leukaemia would account for the most significant savings. Discussion: Despite its limitations, this study provides the first Italian evaluation of the financial impact of rituximab biosimilars and also incorporates the effects of biosimilars on the pricing strategies of the originator (dynamic impact). This dynamic effect is more relevant than the impact of the treatment shift from the originator to biosimilars. Our hope is that these savings will be used to cover new cost-effective drugs and not just for cost-cutting policies

    RSM and Artificial Neural Networking based production optimization of sustainable Cotton bio-lubricant and evaluation of its lubricity &amp; tribological properties

    Depletion of mineral reservoirs along with health and environmental concerns have led to a greater focus on bio-lubricants. The purpose of this study was to analyze and optimize the reaction conditions of the transesterification process for cotton biolubricant synthesis by using Response Surface Methodology (RSM). In RSM, Rotatable central composite design was selected to examine the effect of reaction input factors on the yield of cotton bio-lubricant during the transesterification process. ANOVA analysis showed that temperature was the most significant factor followed by time, pressure and catalyst-concentration. Optimum reaction conditions obtained by RSM for maximum TMP tri-ester (cotton bio-lubricant) yield of about 37.52% were 144 °C temperature, 10 h time, 25 mbar pressure, and 0.8% catalyst-concentration. RSM predicted results were successfully validated experimentally and by artificial neural networking. About 90%–94% cotton seed oil bio-lubricant was obtained after purification and its physiochemical, lubricity and tribological properties were evaluated and found comparable with ISO VG-46 and SAE-40 mineral lubricant. Hence, cottonseed oil is a potential source for the bio-lubricant industry

    Behavioural signs of pain in cats: an expert consensus

    To identify where a consensus can be reached between veterinary experts in feline medicine on the core signs sufficient for pain (sufficient to indicate pain when they occur, but not necessarily present in all painful conditions) and necessary for pain (necessary in the presence of pain, but not always indicative of pain). Methods A modified Delphi technique was used, consisting of four rounds of questions and evaluation using nineteen participants during the period December 2014 and May 2015. Agreement was considered to be established when 80% of the experts concurred with the same opinion. Results Twenty-five signs were considered sufficient to indicate pain, but no single sign was considered necessary for it. Discussion Further studies are needed to evaluate the validity of these 25 behavioural signs if a specific pain assessment tool is to be developed that is capable of assessing pain in cats based on observational methods alone. The signs reported here may nonetheless help both vets and owners form an initial evaluation of the pain status of cats in their care

    Sulfate Activation in Mitosomes Plays an Important Role in the Proliferation of Entamoeba histolytica

    Mitochondrion-related organelles, mitosomes and hydrogenosomes, are found in a phylogenetically broad range of organisms. Their components and functions are highly diverse. We have previously shown that mitosomes of the anaerobic/microaerophilic intestinal protozoan parasite Entamoeba histolytica have uniquely evolved and compartmentalized a sulfate activation pathway. Although this confined metabolic pathway is the major function in E. histolytica mitosomes, their physiological role remains unknown. In this study, we examined the phenotypes of the parasites in which genes involved in the mitosome functions were suppressed by gene silencing, and showed that sulfate activation in mitosomes is important for sulfolipid synthesis and cell proliferation. We also demonstrated that both Cpn60 and unusual mitochondrial ADP/ATP transporter (mitochondria carrier family, MCF) are important for the mitosome functions. Immunoelectron microscopy demonstrated that the enzymes involved in sulfate activation, Cpn60, and mitochondrial carrier family were differentially distributed within the electron dense, double membrane-bounded organelles. The importance and topology of the components in E. histolytica mitosomes reinforce the notion that they are not “rudimentary” or “residual” mitochondria, but represent a uniquely evolved crucial organelle in E. histolytica

    Does left atrial volume affect exercise capacity of heart transplant recipients?

    <p>Abstract</p> <p>Background</p> <p>Heart transplant (HT) recipients demonstrate limited exercise capacity compared to normal patients, very likely for multiple reasons. In this study we hypothesized that left atrial volume (LAV), which is known to predict exercise capacity in patients with various cardiac pathologies including heart failure and hypertrophic cardiomyopathy is associated with limited exercise capacity of HT recipients.</p> <p>Methods</p> <p>We analyzed 50 patients [age 57 ±2 (SEM), 12 females] who had a post-HT echocardiography and cardiopulmonary exercise test (CPX) within 9 weeks time at clinic follow up. The change in LAV (ΔLAV) was also computed as the difference in LAV from the preceding one-year to the study echocardiogram. Correlations among the measured parameters were assessed with a Pearson's correlation analysis.</p> <p>Results</p> <p>LAV (n = 50) and ΔLAV (n = 40) indexed to body surface area were 40.6 ± 11.5 ml·m<sup>-2 </sup>and 1.9 ± 8.5 ml·m<sup>-2·</sup>year<sup>-1</sup>, data are mean ± SD, respectively. Indexed LAV and ΔLAV were both significantly correlated with the ventilatory efficiency, assessed by the VE/VCO<sub>2 </sub>slope (r = 0.300, p = 0.038; r = 0.484, p = 0.002, respectively). LAV showed a significant correlation with peak oxygen consumption (r = -0.328, p = 0.020).</p> <p>Conclusions</p> <p>Although our study is limited by a retrospective study design and relatively small number of patients, our findings suggest that enlarged LAV and increasing change in LAV is associated with the diminished exercise capacity in HT recipients and warrants further investigation to better elucidate this relationship.</p
